Summary:

- Experimental work with cell lines: HeLa, A549 (adenocarcinomic human alveolar basal epithelial cells ), SW620 (colon carcinoma cell lines), HepG2 (liver hepatocellular cells), HEK (Human Embryonic Kidney 293 cells), MCF-7 cells (human breast adenocarcinoma cell line)
- Planning of experiments
- Electroporation of cancer cell lines with a various reporter gene constructs based on secreted alkaline phosphatase SEAP, Green Fluorescent Protein (GFP), Luc Reporter Gene System, Red Fluorescent Protein (RFP), Enhanced Green Fluorescent Protein (EGFP), by using Cellaxess HT electroporation module – making relevant protocols with different pulses and voltages – adjusted to the type of cells and their viability.
- Supervisor of the Cellaxess HT electroporation platform, including detailed maintenance, care of spare parts and a fluent technical efficiency
- Treatment of cell lines after an electroporation with RedDot1, RedDot2, TNFα, PMA
- Transformation of competent cells Top10 and DH10B with a ready constructs from Gene Copoeia and Switch Gear (for example - GAPDH, EF1A, S 714561 – ABCG2, S 715295- ABCC4, S715931 – ABCB1, HPRM 24492 – ABCG2, ABCB10 3UTR_01 and many others) for a further experiments based on electroporation with Cellaxess HT platform
- Knowing of cloning procedure of interesting genes promotores into vectors with codon usage for high expression – mostly into the p TRP_PRL vector; (projecting primers for PCR reaction, familiar with programs Thermo Calc, Oligo3, Ape; using of restriction enzymes (Thermo Scientific, Fermentas); further ligation (T4 ligase).
- Isolating endotoxin-free plasmid DNA from recombinant E. coli cultures (Sigma Aldrich Gen Elute HP Endotoxin – Free Plasmid Maxiprep Kit) and also isolating high-quality plasmid DNA for use in eukaryotic transfection and in vitro expression experiments (Promega Pure Yield Plasmid Miniprep System)
- Confocal microscopy (Zeiss LSM 780 system)
